No, they block that specific email address from being used for a while (over 6 months, I believe). That's so someone else doesn't get to it (before you) and start getting your mail by accident from folks that don't know you shut it down.

Yes... One click on the spam button probably won't stop such a person or mail. But... when others receive the same mail and also click the spam button the spam rating of that person / message gets higher until it is officially considered spam...
